The Stem Cell Analytics Core provides technical assistance and advice on a range of issues including:

  • Stem cell culture
  • Experimental design
  • Assay development and data analysis
  • Pluripotency testing
  • Sorting and plating of stem cells for experimental analysis
  • New cell line development (from frozen stocks and primary material)
  • Media testing

Upon request, the Stem Cell Core can provide protocols including:

  • Media preparation
  • MEF derivation, culture and cryopreservation
  • Feeder layer production
  • hESC culture (enzymatic and manual)
  • Feeder free hESC culture
  • Cryopreservation of hESC’s
  • Quality control analysis

This manual is available to all clients but intended for researchers who are new to the field of Stem Cell Research.

2.5 Day Human Pluripotent Stem Cell Workshop

The Children's Hospital Los Angeles Stem Cell Analytics Core hPSC workshop is an introductory two and a half day course in human pluripotent stem cell (hPSC) techniques aimed at researchers who are interested in performing experiments with hPSC.

Covered topics include:

  • Preparation of MEF feeder plates
  • Preparation of Matrigel coated plates
  • Feeder free culture (mTeSR1, StemPro and Matrigel/MEF-CM)
  • Enzymatic passaging ontp MEF feeders
  • Manual passaging onto MEF feeders
  • Cryopreservation of hPSC by slow freezing
  • Thawing hPSC
  • Setting up an hPSC lab
  • Quality control testing hPSC cultures
  • Production of embryoid bodies

5-Day Comprehensive hPSC Techniques Course

The USC Stem Cell Core offers a comprehensive 5 day course in human pluripotent stem cell (hPSC) culture techniques. This course is designed for researchers and post graduate students who are interested in establishing a hPSC laboratory and transitioning into the hPSC field.

Covered topics include:

  • Media production (regular growth media and MEF-CM)
  • Matrigel coating
  • hPSC subculture (manual and enzymatic)
  • Culture of ESI serum adapted hESC lines
  • Feeder free hPSC culture (mTeSR1, StemPro and MEF-CM)
  • Techniques for plating dissociated hPSC's
  • EB production (via scraping and centrifugation
  •  Primary MEF isolation
  • Fibroblast culture
  • MEF Feeder layer production
  • Quality control testing hPSC cultures
