The Vector Core provides assistance in the design and construction viral vectors, as well as in the production of viral particles for gene-modified in vitro as well as in vivo animal studies. The Vector Core also provides consulting services to help researchers identify their gene transfer needs, determine the best way to produce gene-modified cells, and to assist in experimental troubleshooting.

Equipment and Software

Hollow fiber tangential flow filtration (TFF) columns (Spectrum Labs) efficiently filter and concentrate lentiviral vectors up to 100 fold with high recovery (>90%). The addition of a second TFF column for smaller volumes and the sequential use of 2 TFF columns enables concentration of up to 5000 mL viral supernatant first to 10-20 mL, and then to 1-2 mL.

This eliminates the need for ultracentrifugation, and results in concentrated virus 2500 fold relative to unfiltered viral supernatant. The final viral titers range from 1 x 10^9 to 1 x 10^10 viral particles per mL.
